Moderna, a clinical-stage biotech that had never brought a product to market, dosed the first volunteer in an NIH-led Phase 1 trial of its mRNA-1273 vaccine on March 16, 2020 — 63 days after the viral sequence was selected. It then made a bet few peers matched: manufacturing at scale before efficacy was proven.

In May 2020 Moderna signed a 10-year collaboration with Lonza to build capacity for up to 1 billion doses a year, saying the deal would accelerate its manufacturing tenfold, with first batches at Lonza US planned for July 2020. BARDA contract funding, announced in April 2020, covered part of the scale-up.

The bet paid off. In November 2020 interim Phase 3 data showed 94.5% efficacy, and the vaccine was authorized in the US. In 2021 Moderna shipped 807 million doses and reported revenue of $18.5 billion — from $803 million in 2020 — and net income of $12.2 billion after a $747 million loss.

The vaccine showed 94.5% efficacy in November 2020; 2021 revenue reached $18.5 billion and net income $12.2 billion, from a $747 million 2020 loss.

Manufacturing lead time, not clinical data, was the pandemic's critical path.

Lonza's scale expertise multiplied Moderna's capacity tenfold.

BARDA funding shared the financial risk of building before approval.

mRNA's platform design made a new vaccine cheap to produce once scaled.

When speed is the whole game, build manufacturing capacity in parallel with clinical proof — the risk of overbuilding beats the cost of arriving late.

Spikevax became one of the most widely used vaccines, approved or authorized in more than 70 countries, and Moderna expanded its mRNA pipeline to 44 programs with a $3 billion buyback. The company it became — a commercial pharmaceutical giant — bore little resemblance to the clinical-stage firm of early 2020.
